Assay specificity The assay is specific for pathogenic Cryptosporidium species and Giardia assemblages A&B. Other related protozoa that were tested (e.g., Entamoeba histolitica, Babesia canis, Babesia gibsoni, Theileria annae, Toxoplasma gondii and Trypanosoma brucei) showed negative results under the same conditions.
Quantification The above is a typical standard plot of serial decimal dilutions of target Cryptosporidium and Giardia DNA sequences (green dots). Blue dots indicate water samples containing 30 and 3 Cysts/Oocysts, respectively. Environmental water samples were spiked with Cysts and Oocysts from validated spiking stocks (Waterborne, Inc.) and tested with AquatiqPCR©-C/G kit. Results The number of Giardia Cysts and Cryptosporidium Oocytes can be deduced by a standard plot based on serial decimal dilutions of target DNA oligo’s. Taking into account that every Giardia Cyst contains 8 sets of genomic DNA and each Cryptosporidium Oocyst contains 4 sets, we are able to translate the DNA copy number from the qPCR reaction into actual numbers of Cyst/Oocyte.
